Creative Proteomics Initiatives New De Novo Sequencing Service

  • It is widely known that the sequence of protein plays an important role in studying the biological function of the peptide/protein. However, it still a very big challenge when it comes to complete characterization of peptides/proteins, including post-translational modifications (PTMs), sequence mutations and variants. Normally two approaches are applied to determine the sequence of peptide/protein by mass spectrometry, which are database search and de novo sequencing. Database search approach compares acquired mass spectra to a database of known protein sequences to identify the protein sequences while De novo sequencing is an approach in which amino acid sequences are directly interpreted from tandem mass spectra without the assistance of a database. However, database search approach does not work if the protein sequence does not exist in the current database. As a result, de novo sequencing is the ideal method for identifying novel peptides, unsequenced organisms, and antibodies drugs.

    At Creative Proteomics, we successfully tackle these challenges by using the following four steps. Firstly, to avoid the false positivity led by low mass resolution and accuracy by fourier transform ion cyclotron resonance mass spectrometry (FTICR-MS), which has the highest mass resolution and accuracy. Secondly, applying both bottom-up and top-down mass spectrometry to analyze the same sample. Thus the peptide/protein sequence can be confirmed in a better way by combing these data. Thirdly, four types of fragmentation echniques are used to provide more fragment ions from the same peptide/protein and these complementarity data will sure to improve the ion assignment. Finally, we also offer chemically derivation to identify ion series.Creative proteomics gives the opportunity to gain a better understanding of the peptide/protein de novo sequencing.

    Creative Proteomics is continuing to optimize analytical software, facilities, and experiment design. A detailed and precise technical report will be provided at the end of the whole project, including the sample preparation, HPLC-MS/MS parameters.
